Abstract

Oxidative stress caused by free radicals can damage cellular components and contribute to various degenerative conditions. The rhizome of Zingiber zerumbet Sm. is known to contain antioxidant compounds, including flavonoids, which have the potential to reduce oxidative stress and inflammation. This study aims to explore the total flavonoid content and antioxidant activity in the fractions of Zingiber zerumbet rhizome as an effort to identify a source of natural antioxidants. The method applied in this study is fractionation through liquid-liquid extraction. This process separates bioactive compounds from the rhizome of elephant ginger using various solvents. Each fraction was analyzed for total flavonoid content using the colorimetric method and antioxidant activity using the DPPH method, which was measured using a UV-Vis spectrophotometer. The fractionation of 70% ethanol extract from the rhizome of Zingiber zerumbet revealed that the aqueous fraction had the highest yield at 76.00%, followed by the ethyl acetate fraction (20.33%), methanol (2.67%), and n-hexane (1.00%). Measurement of total flavonoid content indicated that the aqueous fraction contained 5.55 mg QE/g of extract, while the methanol fraction was slightly higher at 5.64 mg QE/g of extract. The ethyl acetate fraction recorded the highest content at 29.25 mg QE/g of extract, whereas the n-hexane fraction showed a content equal to that of the aqueous fraction, at 5.55 mg QE/g of extract. Antioxidant activity tests demonstrated that the ethyl acetate fraction had an IC₅₀ value of 17.11 µg/mL, categorizing it as very strong. The methanol and aqueous fractions had IC₅₀ values of 53.55 µg/mL and 64.44 µg/mL, respectively, which are classified as strong, while the n-hexane fraction exhibited no antioxidant activity with an IC₅₀ value of 905.55 µg/mL. In conclusion, the water fraction of 70% ethanol extract of elephant galangal rhizome had the highest yield and the ethyl acetate fraction had the highest total flavonoid content and the strongest antioxidant activity.
