Abstract

Lempuyang gajah is a member of the Zingibereace family that is often used in Indonesia as a traditional herbal medicine. Lempuyang gajah, especially the rhizome, is used in traditional medicine because it has a number of benefits based on its biological and pharmacological characteristics. Lempuyang gajah rhizome extract contains many secondary metabolite compounds, including alkaloids, saponins, flavonoids, and polyphenols, as well as terpenoids and bioactive compounds, namely curcumin. This study aims to identify the compound content of the fractions (water, metanol, ethyl acetate, and n-hexane) of ethanol extract of Lempuyang gajah rhizome using phytochemical screening and TLC methods. This study is an experimental study with a qualitative approach with a one-shot case study research design. Fractions of ethanol of lempuyang gajah rhizome were obtained by liquid-liquid extraction method. The fractions obtained were subjected to phytochemical screening and further confirmed by TLC method. The results showed that the water fraction was the highest yield, which was 2.42 g with a percentage of 48.52% of the total yield weight. In the phytochemical screening of the ethyl acetate fraction of the Lempuyang gajah galangal rhizome (Zingiber zerumbet) contained saponins, polyphenols, flavonoids, triterpenoids, and quinones. The methanol fraction contained flavonoids and quinones. And the water fraction contained saponins and tannins. While the n-hexane fraction did not contain any compounds. The TLC test results showed that the methanol fraction of the Lempuyang gajah galangal rhizome contained curcumin with an Rf value identical to the Rf value of the comparator, which was 0.88. The results of phytochemical screening of the ethyl acetate fraction contained the most secondary metabolites, while the TLC results of the metabol fraction showed the presence of curcumin compounds.
